Abstract:Abstract: The seeds of Gleditsia vestita Chun et How ex B. G. Li were used as explant material. Through callus differentiation, adventitious bud multiplication, acclimatization and transplantation, the tissue culture system was set up. The results are as follows: 1) The best group appeared in the medium containing MS+6-BA 3.0 mg/L+KT 0.4 mg/L+IAA 0.3 mg/L, in which the callus and differentiated adventitious buds were induced synchronously from the cotyledons and hypocotyls. The callus induction rate reached 90.5% and synchronous differentiation rate reached 77.9%; 2) The average proliferation times of buds were 4.0 in the medium containing 1/2MS+6-BA 1.0 mg/L+IBA 0.3 mg/L+NAA 0.2 mg/L; 3) The rooting rate was 100% in MS+NAA 0.5 mg/L+IAA 0.25 mg/L. The plantlets grew well with 90% survival after transplantation.
