Abstract:Abstract: To explore the effect of salvianic acid on the cell phenotype and partial immune functions of mouse bone marrow-derived dendritic cells (DCs), mouse bone marrow cells were separated and culutred in vitro firstly, and were then induced to differentiate into DCs by IL-4 and GM-CSF. The percentage, MHC-II, CD80 and CD86 expression levels and phagocytosis of these DCs were detected by flow cytometry before and after treatment with salvianic acid A. The cytokine concentrations in the culture supernatant of DCs co-cultured with T cells were examined by ELISA. Results showed that the percentage of DCs and the number of FITC-Dextran positive cells were obviously increased, while the ratio of MHC-IIhigh/MHC-IIlow, the mean flourescence intensity of MHC-IIhigh and MHC-IIlow, and the expression levels of CD80 and CD86 on DCs were decreased significantly after treatment with salvianic acid A. In addition, salvianic acid A enhanced the secretion ability of IL-10, and inhibited INF-γ secretion ability by T cells stimulated by DCs. These results indicate that salvianic acid A could induce bone marrow cells to differentiate into DCs, but inhibit DC maturation and its antigen presenting funcion, resulting in decreased expression of surface marker on DCs. Besides, salvianic acid A could strengthen the ability of antigen uptake and phagocytosis of DCs, and alleviate the inflammatory reaction mediated by DC and T cell interactions. These results will provide strong support for the administration of salvianic acid A on the immune diseases induced by T cell excessive activation.
