Abstract:Abstract: To investigate the roles of amyloid precursor protein (APP) in nasopharyngeal carcinoma CNE2 cells, siRNA expressing plasmids were transfected into CNE2 cells by Lipofectamine 2000 reagent, and a non-specific plasmid was used as control. Immunofluorescence and Western-blot were used to detect the transfection efficiency and the expression level of APP protein, respectively. MTT test and wound healing assay were performed to evaluate the proliferation and migration abilities of CNE2 cells. The results showed that APP protein was significantly lower in the interference group than that in the control group (P<0.05). Interference with APP gene could inhibit the proliferation and reduce the healing of scratches of CNE2 cells significantly. These results suggested that RNAi could effectively inhibit the expression of APP protein, and that proliferation and migration of CNE2 cells could be suppressed by inhibiting APP expression. The study provides a new clue for gene therapy on nasopharyngeal carcinoma.
