Abstract:KIAA0146 is a scaffold protein of homologous recombinant protein complex BLM/RAD51. Currently, there are few KIAA0146-related experimental studies. The effective bioinformatics analyses were therefore performed for providing clues for further experimental study of KIAA0146. Several transcription start sites and promoters were found at the KIAA0146 gene. Besides, two CpG islands were found on the flank of promoter (2 127~2 177 bp) and (3 127~3 177 bp), respectively. It was found that KIAA0146 was an unstable hydrophilic protein without signal peptide and transmembrane domain. It was therefore considered as neither a secreted protein nor a transmembrane protein. However, the KIAA0146 was considered as nuclear protein with nuclear localization signal “RARGSKRKR” at its N-terminal. The constructed 3D structure of KIAA0146 showed that it contained a nucleic acid-binding domain. It was finally found that RAD51, ATM, RAD51B, MRE11A, BLM, DNA2, RMI1, BRCA1, BRCA2 and XRCC2 might interact with KIAA0146. In addition, KIAA0146 was considered to involve into the biological processes including DNA damage response and repair, development and cell metabolism regulation through these interacting proteins. In conclusion, the analyses of gene structure, protein subcellular location, 3D structure and potential molecular function of KIAA0146 provide important theories and new clues for further experimental study on this molecule.
