Abstract:To investigate the effect of transfer of peroxiredoxin Ⅱ (Prx Ⅱ) from membrane to cytoplasm on erythrocyte osmotic fragility under oxidative stress, the change of erythrocyte osmotic fragility induced by H2O2 was examined, and the condition of Prx Ⅱ transfer from membrane to cytoplasm in erythrocytes and the expression of the membrane proteins band 3 and spectrin of red blood cells were detected by Western-blot. The results showed that the osmotic fragility of red blood cells was increased, and Prx Ⅱ was transferred from the erythrocyte membrane to cytoplasm during oxidative stress. At the same time, the expression of proteins that maintain the stability of red blood cell membrane and the integrity of the cytoskeletal structure and function was reduced. These results indicated that Prx Ⅱ in the erythrocytes could be transferred from the membrane to cytoplasm under oxidative stress, and this would lead to the decreased expression levels of band 3 and spectrin and the increased osmotic fragility of the red blood cells.
