Abstract:Abstract: Obstructive jaundice (OJ) is a common clinical disease. Although the obstruction could be removed by surgery, the resulting liver damage is difficult to repair. Huwentoxin-XI (HWTX-XI) is an effective trypsin inhibitor, but its potential effect on liver injury has not been explored experimentally. On the other hand, aerobic exercise has been reported to be effective in the amelioration of inflammation, and to promote the process of liver regeneration. In order to investigate the antagonistic effect of HWTX-XI treatment combined with aerobic exercise on liver injury caused by obstructive jaundice, a mouse model with the common bile duct hanging at the abdominal wall was established. In addition, treadmill training was adopted for aerobic exercise and HWTX-XI was administrated by tail vein injection. After the intervention, the morphological changes in liver were observed by HE staining and the expression levels of the inflammatory-related molecules were detected by RT-PCR and immunohistochemistry. The results showed that HWTX-XI administration combined with aerobic exercise could be beneficial to the recovery of damaged mouse liver induced by biliary obstructive jaundice. The underlying mechanism could be involved in the down-regulation of factors in the NF-κB signaling. Moreover, the joint effect of HWTX-XI treatment and aerobic exercise was much more efficient than any single-factor intervention.
