Abstract:Abstract: To identify the key genes of esophageal squamous cell carcinoma by bioinformatics and analyze the related biological pathways, the gene chip expression profiles of GSE20347, GSE29001, GSE33426, GSE45168 and GSE70409 were downloaded from GEO database, and the differentially expressed genes were obtained by RobustRankAggreg method. DAVID database was used to annotate the identified genes and ana-lyze the enrichment of KEGG pathway. The protein interaction network recognition module of differentially expressed genes was constructed by STRING database. Finally, based on the software Cytoscape, four algo-rithms including Degree, MCC, MNC and EPC were used to identify the key node genes, and the key node genes were verified in GEPIA database. A total of 373 differentially expressed genes were identified, inclu-ding 154 up-regulated genes and 219 down-regulated genes. These genes were mainly involved in extracel-lular matrix organization, cytoplasm, protein binding, oxidation-reduction process, extracellular exosome, cal-cium ion binding, amoebiasis, PI3K-Akt signaling pathway, ECM-receptor interaction, focal adhesion, and protein digestion and absorption. Two main functional modules and seven key genes CDK1, KIF20A, TTK, CDC45, CCNB2, TPX2 and KIF4A were obtained by protein interaction network and Venn diagram. The re-sults showed that the expression of 7 key genes was increased in esophageal squamous cell carcinoma. There is still less research on KIF20A and KIF4A related to esophageal squamous cell carcinoma. The key genes and biological pathways obtained in this study may provide bioinformatics information for the diagnosis and treatment of esophageal squamous cell carcinoma.
