Abstract:Abstract: In order to excavate the bHLH transcription factors regulating gentiopicroside biosynthesis in Gentiana rigescens, gene sequence of transcription factor CrBIS1 (bHLH iridoid synthesis 1) controlling iridoid biosynthesis in Catharanthus roseus was aligned against transcriptome database of G. rigescens, and then a candidate regulatory gene GrbHLH1 was obtained. Based on this sequence, GrbHLH1 gene was cloned from young leaves of G. rigescens by reverse transcription-polymerase chain reaction (RT-PCR), and its bioinformatics analysis and tissue specific expression were analyzed. Results showed that the open reading frame of GrbHLH1 gene (GenBank accession number: KF941186.2) is 1 104 bp and encodes 367 amino acids, mainly expressed in leaves. The relative molecular mass of GrbHLH1 protein is 40.60 kD and its theoretical pI is 4.82. It may be localized in the nucleus and is a hydrophilic and unstable protein without signal peptides. It consists of α-helix (18.53%) and loop (70.30%) with a conserved domain of HLH, belonging to IVa-type bHLH transcription factor. It has the closest relationship with CrBIS1 and CTBIS2 proteins in C. roseus. In summary, our data suggested that GrbHLH1 gene should be a candidate regulatory gene for gentiopicroside biosynthesis, and this lays a foundation for further study on the regulatory role of GrbHLH1 gene.
