Abstract:Abstract: The origin and migration of primordial germ cells (PGCs) have been studied in a variety of fish species, but the visualization and migration of PGCs in polyploid fish have not been reported. The unique fish lineages in our laboratory (including allotetraploid carp, red crucian carp and triploid crucian carp), which have clear genetic backgrounds, were used to label PGCs. In this paper, the mRNA generated by fusion of nanos1-3′-UTR and GFP from zebrafish was injected into the fertilized eggs of polyploid fish, and the PGCs of polyploid fish were labeled for the first time to observe their migration route to the genital ridge. The results showed that the zebrafish nanos1-3′-UTR could label the PGCs of polyploid fish and their migration was similar to those of zebrafish. These results provide some basic data for the study of the origin and migration of PGCs of polyploid fish.
