Abstract��Abstract: In order to solve the low efficiency in lenvivirus packaging and infection, the condition of packaging and infection of lentivirus from the perspective of culture solution pH was optimized. To a lentivirus plasmid vector with copGFP, the calcium phosphate and Lipofectamine2000 methods to transfect 293T cells cultured in different pH culture solution were applied. By fluorescent microscope to compare transfection rate of GFP positive cells, it was found that the calcium phosphate method get high transfection efficiency only in pH (6.5~7.5). For Lipofectamine2000, culture solution pH (6~8) almost had no effect in the transfection rate. When using culture solution of standard pH (7.2~7.4) to package lentivirus and adjusting lenvivirus supernatant collection to different pH and then to infect 293T cells with Polybrene or not, it was found that when pH of lentivirus supernatant collection was near 8~9, the infection rate was the highest, and using Polybrene at the same time help to improve the efficiency of infection.
