Abstract:Abstract: Alzheimer’s disease (AD) is a neurodegenerative disease caused by multiple factors and clinically manifested as irreversible cognitive decline. It has become a severe challenge all over the world. There are two types of AD: familial and sporadic, and more than 95% of all AD cases are sporadic. At present, most studies on AD pathogenesis and drug development are carried out using transgenic animal models. These models are based on genetic mutations that occur in familial AD cases, which only represent the cases asso-ciated with genetic AD, but ignore the critical roles of acquired factors in the disease development. Based on the mechanism hypothesis of AD, this review summarizes the recently developed non-transgenic animal models, mainly mouse and rat models, and discusses their advantages, disadvantages and application prospects, hoping to provide help for researchers in choosing suitable animal models.
