Abstract:Abstract: Nuclear receptors play an important role in the maintenance of cell homeostasis and the occur-rence and development of diseases. In order to explore the role and mechanism of nuclear receptor subfamily 6 group A member 1 (NR6A1) in liver cancer, the abnormally high expression of NR6A1 in liver cancer was analyzed by using The Cancer Genome Atlas (TCGA) and other databases. By Cell Counting Kit-8 (CCK-8), 5-ethynyl-2′-deoxyuridine (EdU) and wound healing assays, it was found that interference of NR6A1 signi-ficantly inhibited the proliferation of liver cancer cells, but did not affect the migration of cells. Through im-munocoprecipitation assay, immunofluorescence staining, RNA interference and overexpression technology, N-myc downstream-regulated gene 1 (NDRG1) was identified as an interacting protein of NR6A1 in liver can-cer. The expression levels of NR6A1 and NDRG1 were positively correlated in liver cancer, and NR6A1 positively regulated the expression of NDRG1. Finally, the results of functional rescue experiment confirmed that interference of NDRG1 could inhibit the increased proliferation caused by NR6A1 overexpression in liver cancer cells. In conclusion, NR6A1 plays an oncogenic role by binding with NDRG1 and upregulating its expression.
