Abstract:Abstract: The toxic effect of carbon tetrachloride (CCl4) combining with diethylnitrosamine (DEN) on kidney was studied. Kunming mice (Mus musculus domesticus) were randomly divided into two groups. The experimental group was injected intraperitoneally with 15% CCl4 in peanut oil twice a week for eight weeks, with free access to water containing 0.07% DEN, and the control group was injected with same dose of peanut oil with free access to purified water. The diet and weight changes of mice were observed. In both experimental and control groups, four mice were killed at weeks 3, 6 and 8, respectively. The kidney tissue was fixed, embedded and sectioned conventionally, and then stained by HE method. The structure and the shape changes of kidney tubules and collecting tubules were observed. The results showed that average mouse weight was significantly higher in the control group than in the experimental group. The structures of kidney in control group were distinct, arranged orderly, while the kidney tissues of experimental group had remarkable degeneration and necrosis and were more and more disorderly. The kidney tubules and collecting tubules showed a dramatic change, with the lumen narrowing down. These results proved that CCl4 has a damage effect on kidney and the damage is more and more serious with the extension of treatment time.
