Abstract:Abstract: To observe the effects of different centrifugal forces on size, RNA and protein of B16- F10 cell-derived extracellular vesicles, centrifugal forces 12 000×g and 100 000×g were used to prepare two types of B16-F10 cell-derived extracellular vesicles. The size of vesicles was analyzed with Nanoparticle Tracking Analysis (NTA), microvesicles were characterized by Western-blot, RNA was analyzed with Fragment AnalyzerTM Automated CE System and proteins were quantified by BCA assay. The average size of vesicles in 12 000×g group was 233±93.5 nm, and that in another group was 175±74 nm (P<0.05). The protein concentration in 12 000×g group was 3.942±0.427 g/L, and that in another group was 2.428±0.113 g/L (P<0.01). Moreover, Tsg101 was determined in 100 000×g group. The RNA concentration in 12 000×g group was 22.662 0 mg/L, and the length of majority fragments was 258 nt; while in another group, the RNA concentration was 3.280 1 mg/L and the length of majority fragments was 283 nt (P<0.05). Therefore, extracellular vesicle size was associated with centrifugal force, and different sizes of vesicles contained diverse proteins and RNAs.
