Abstract:Abstract: The key of fish artificial gynogenesis is to know the time of the second polar body release and the first mitosis. Fertilization cytology is the effective method. Crossbreeding between Epinephelus lanceolatus (♂ ) and Epinephelus coioides (♀) was conducted by artificial fertilization. The fertilized eggs were fixed in Smith’s solution at different time points after fertilization, embedded by paraffin, sectioned, stained by H. E. and observed with microscope. The results showed fertilization procedure. The Epinephelus lanceolatus sperm entered the Epinephelus coioides eggs at 30~60 s post-fertilization. Sperm-aster appeared at 3 min post-fertilization. The second polar body released at 5 min post-fertilization. Male pronucleus was formed prior the formation of female pronucleus, and later they got closer to each other at 7~15 min post-fertilization. First karyokinetic division was developed at 15 min post-fertilization. The time of Epinephelus lanceolatus sperm entering the Epinephelus coioides eggs had no significant difference to the other teleost fish; however, the time of the second polar body release and the first mitosis was earlier than the other fish. The results showed basic fertilization procedure between Epinepphelus lanceolatus and Epinephelus coioides, could provide the theory foundation for artificial gynogenesi.
