Abstract:Abstract: To explore the effect and molecular mechanism of miR-448 in human lung cancer proliferation and migration. The miR-448 expression level was detected by RT-qPCR. The miR-448 mimic and inhibitor were transfected into human lung cancer A549 cell line. MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazoliumbromide), colony formation and transwell were performed to evaluate the ability of lung cancer proliferation and migration. Western blot was performed to detect the EMT (epithelial-mesenchymal transition) markers expression. RT-qPCR was used to detect the EMT-related transcription factor mRNA expression. The miR-448 expression level was down-regulated in primary lung cancer tissues compared with the adjacent normal lung tissue by RT-qPCR. Overexpression of miR-448 inhibited cell proliferation and migration, whereas suppression of miR-448 promotes cell proliferation and migration in A549 cell line by MTT, colony formation and transwell assays. Moreover, the miR-448-reduced cells showed the decreased epithelial marker E-cadherin and increased mesenchymal marker Vimentin by Western blot. Furthermore, the mRNA expression levels of EMT-related transcription factor Twist1 and ZEB1 were down-regulated in miR-448-reduced A549 cells. miR-448 may inhibit cell proliferation, migration in lung cancer A549 cells through suppression of EMT phenomenon.
