Abstract:Abstract: Previous studies have found that increasing multiplicity of infection (MOI) can effectively improve the efficiency of adenoviruse serotype 5 (Ad5) infection in T cells; however, little is known about the underlying mechanism. Here, Jurkat T cells were selected as target cells, and the effects of different MOI on the viral binding and viral entry in the replication cycle of Ad5-GFP were investigated by means of quantitative fluorescence PCR and transmission electron microscopy. It was found that high MOI significantly increased the number of adenovirus into the T cell during the endocytosis process, but not the number of adenovirus adsorbed on T cells. These results indicate that increase of MOI is beneficial to the adenovirus into the T cells by endocytosis, improving the efficiency of adenovirus infection in T lymphocytes, and this provide a theoretical basis for further study of the adenovirus infection mechanism in T lymphocytes.
