Abstract:Abstract: Zinc finger and BTB domain containing 40 (ZBTB40) is a member of ZBTB transcription factor family. The tissue expression pattern and molecular functions of ZBTB40 in mammal are still unclear. In or-der to investigate the expression pattern and physiological function of the ZBTB40 gene, the relative abun-dance of ZBTB40 mRNA and protein in eight human cell lines and nine mouse tissues was detected. The re-sults showed that ZBTB40 expressed in most of the human cell lines (6/8) and three kinds of mouse tissues (liver, sternum and testis). The ZBTB40 abundance in mouse liver was significantly higher than that in other tis-sues, suggesting that ZBTB40 may be important for physiological functions of liver. To validate the hypothe-sis, the level of ZBTB40 in five pairs of human hepatocellular carcinoma (HCC) and adjacent tissues was exa-mined. The results showed that ZBTB40 was highly expressed in the nucleus of liver cancer cells, but the expression in carcinoma stromal cells was the lowest. Survival analysis suggested that the expression of ZBTB40 was negatively associated with the overall survival time of HCC patients, indicating that it may be a potential prognostic biomarker. In addition, the viral expression plasmid of ZBTB40 gene was constructed, and cell lines stably expressing human ZBTB40 were obtained after U2OS cells were infected, which laid a foundation for further molecular mechanism and functional research.
