Abstract:Abstract: Collagens are the main component of extracellular matrix. In pathological environments, such as trauma or burn, collagen degrades or denatures into gelatin, and macrophages are activated and recruited to the lesion to mediate the inflammatory response. Collagen fragments can mediate inflammatory responses. Gelatin is derived from collagen denaturation or its fragments. Now there are only a few studies on gelatin-as-sociated inflammatory responses. In the present study, the effects of gelatin on RAW264.7 mouse macrophage cells were explored. The morphological changes, viability and proinflammatory cytokine production of the cells were detected. The results showed that gelatin promoted RAW264.7 cell aggregation in a concentra-tion- and time-dependent manner, but did not influence cell viability. Meantime, gelatin-coated plates stimu-lated production of proinflammatory cytokines in RAW264.7 cells. Furthermore, it was found that gelatin acted on RAW264.7 cells through NF-κB signaling pathway. The results suggested that gelatin may play an important role in macrophage-mediated inflammatory response.
