摘要

摘 要: 高尿酸血症(hyperuricemia, HUA)是心脏和肾脏疾病发生与发展的重要危险因素, 降尿酸治疗是应对HUA的首要处理方案。课题组前期发现, 天然产物黄芩素(baicalein, BAL)具有降尿酸作用。本研究使用氧嗪酸钾联合次黄嘌呤构建持续8周的慢性HUA小鼠模型, 考察BAL对心脏和肾脏的影响, 实验过程中使用别嘌醇(allopurinol, AP)作为阳性对照药物。结果显示, BAL可以改善HUA下的心肌肥厚、心肌细胞排列紊乱及心肌间质纤维化等现象, 其趋势与AP一致; BAL无法改善HUA造成的肾功能下降, 以及肾小球基膜增厚、肾小管扩张等现象, 与AP相悖。综上所述, BAL在慢性HUA下对心脏具有保护作用, 而对肾脏作用不显著。本研究为BAL在HUA合并心肾疾病的研究提供了有效信息。

Abstract

Abstract: Hyperuricemia (HUA) is an important risk factor for the incidence and development of heart and kidney diseases. Urate-lowering therapy is the primary treatment for HUA. We previously discovered natural product baicalein (BAL) showing urate-lowering effect. Herein, effects of BAL on heart and kidney were as-sessed in a chronic HUA mouse model induced by co-administration of potassium oxonate and hypoxanthine for 8 weeks. Mice treated with allopurinol (AP) were used as positive control. The results showed that BAL ameliorated HUA associated cardiac hypertrophy, myocardial fiber disarray and interstitial fibrosis, which was consistent with AP in terms of efficacy. However, unlike AP, BAL didn’t ameliorate HUA associated re-nal function decline, glomerular basement membrane thickening and tubular dilation. It indicated that BAL may protect heart in HUA while not contribute to kidney protection. This study may provide useful informa-tion for BAL application in research of HUA with heart or kidney diseases.
