Abstract:Abstract: To explore the expression level and clinical significance of transcription elongation factor B subunit 1 (TCEB1) gene in hepatocellular carcinoma (HCC) by bioinformatic analysis, GEPIA and UALCAN were used to analyze the gene expression and its relationship with clinicopathological characteristics and prognosis of the patients, and the cBioPortal and LinkedOmics databases were used to analyze its changes and correlated genes in HCC, respectively. GO analysis and KEGG pathway enrichment analysis of TCEB1-related genes were performed by Metascape. Finally, the protein-protein interaction network was constructed by GeneMA-NIA. Analysis using GEPIA and UALCAN showed that TCEB1 was highly expressed in HCC tissues (P<0.05), and the higher the mRNA expression level, the worse the tumor grade and prognosis (P<0.05). The cBioPor-tal database showed that 5% (52/1 136) HCC samples had TCEB1 alteration, and the copy number amplifi-cation was positively correlated with its mRNA expression. The patients with TCEB1 alteration had a signifi-cantly poorer prognosis (P=0.012 9). GO analysis showed that the molecular functions of TCEB1 and correla-ted genes were mainly related to transcription factor binding, ubiquitin-like protein transferase activity, and cadherin binding. GeneMANIA analysis showed that TCEB1 interacts with TCEB2, VHL, CUL2 and other proteins. The above results suggest that TCEB1 may serve as a biomarker for the prognosis of HCC patients and a new potential target for HCC treatment.
