Abstract:Abstract: Rv1057 is the only propeller protein in Mycobacterium tuberculosis, but its biological functions are still unclear. Here, the effects of Rv1057 gene on immune response of macrophages in the early infection with M. tuberculosis were studied. The macrophages were infected by Rv1057 deletion strain D1057 and the wild type strain H37Rv, respectively, then the multiplication rate of M. tuberculosis in macrophages was detected. Finally, the cytokine expression levels in the early stage of macrophage infection were analyzed. The live M. tuberculosis and its multiplication rate in macrophages infected with D1057 were significantly lower than those with H37Rv. The expression levels of IL-1β, IL-10, TNF-α and IFN-γ were obviously decreased in macrophage infected by D1057. However, the expression level of IL-8 was increased, but not obviously. The experiments proved that Rv1057 deletion could reduce the immunogenicity of M. tuberculosis, and that Rv1057 was involved in the immune response between macrophages and M. tuberculosis. The experiments provide a foundation for further study in biological functions of Rv1057 and pathogenic mechanisms of M. tuberculosis.
